As the name says, 'animation cancel' is practically when you cancel your champs animation. This is usually referred to when you cancel your auto-attacks. Take example Ashe, you target a minion and start to attack it using auto-attack, but then you realize 'Oh, the creep ain't low enough (HP wise)' and so you decided to cancel your attack, but your Ashe's auto-attack has been released and then you :
1. moved your Ashe to some place (whatever is the place) OR
2. target another minion before your projectile/attack reaches that previous minion
Option 1 is trickier to do while option 2 is much easier to do (well least for me). The term 'animation cancel' was used in DoTA frequently when mentioning about chasing your enemy while decreasing the delay of your auto-attack. By doing 'animation cancel' you cut off the time it takes for your Champ to do another attack thus causing your Champ to do the next attack in quick succession.
In LoL it's much much harder to do 'animation cancel' compared to in DoTA (system? skill? graphic? I seriously don't know why). And even if you do an 'animation cancel' it is most likely that your target didn't receive the first hit (in LoL). You may have done it without thinking, like for example when you attack a low-health enemy beside his/her tower while you also have low health too. Because you're afraid of the danger you might die you quickly click your enemy and immediately clicked the ground causing an 'animation cancel'.
As far as my memory goes, I don't think there's animation cancelling for spells but I'm not sure. Hope this helps
